PCR 결과로 그래프 만들기

① housekeeping gene을 넣은 그룹을 기준으로 잡는다.

(프로그램 설정 시 Reference sample은 negative control sample, endogenous control은 housekeeping gene을 넣는다)

 

② housekeeping gene의 CT값을 사용해 각 그룹의 CT값에서 dCT를 구한다.

dCT= CT(gene)-CT(B-actin 같은 control)

 

③ housekeeping gene을 넣은 control 그룹의 값을 뺀다.

ddCT= dCT (실험군) - dCT (negative control, 여기서는 MSC)

실험 그룹은 N.C 그룹보다 CT값이 작으므로 -값이, housekeeping gene 그룹은 0값이 나온다.

 

④ 2^(-ddCt)(=RQ)의 값을 구한다(이때 housekeeping gene 그룹의 값은 0이므로 2^(-ddCt)의 값은 1이 된다)

 

⑤ RQ값을 이용해 그래프를 그린다.
